Abstract
| - An ultrahigh-throughput method for acquiring 1H NMRspectra is described. By constructing a continuous flowsystem utilizing an HPLC pump, autosampler, and acapillary flow NMR probe, it was possible to inject samplesinto the NMR spectrometer every 30 s using a continuousflow rate of 30 μL/min. 1H NMR spectroscopic data wereacquired continuously into a pseudo-2D data file, with a96-well-plate completed in <50 min. Spectra in continuous flow mode were readily obtained from ∼3.4 μg (500MHz), while the LOD was <850 ng. There was found tobe little variation in either sample broadening within theflow system or signal intensities between multiple injections. This system offers several advantages over moreconventional NMR spectroscopic analyses, notably thelimited solvent required, high sensitivity, high speed, andimproved spectral quality as a result of reduced spectral“dead” regions resulting from residual solvent levels.
