Abstract
| - The amino terminus of mouse epidermal growth factor (mEGF) was coupleddirectly to the aldehydeend of dextran through a reductive amination procedure. Thehighest coupling efficiency was ∼80%and could be reached after ∼24 h of reaction time at pH 8. Gelfiltration on Sephadex G-50 Fineremoved free mEGF from the conjugate. Preparative polyacrylamidegel electrophoresis was used toseparate the conjugate from excess noncharged dextran. Theconjugate bound specifically to the EGFreceptor on cultured glioma cells as shown in displacement tests withfree mEGF. The conjugatewas stable in the pH interval 4−9, in 2 M sodium chloride, in 7 Murea, and in human serum andcould still bind to the EGF receptor after such treatments. Theconjugates are candidates for targetednuclide therapy.
