Abstract
| - The systemic bioavailability of polynuclear aromatichydrocarbons (PAH) from ingested soils containingmanufactured gas plant (MGP) tar was evaluated in mice.Soil and organic extract of each soil were incorporatedinto a diet and fed to mice for two weeks. 1-Hydroxypyrenelevels in urine and chemical:DNA adduct levels in lungswere used as biomarkers of PAH systemic bioavailability.Estimates of PAH relative bioavailability were determinedby comparing the bioavailability observed between each soiland corresponding organic extract. In all but one case,bioavailability estimates based on 1-hydroxypyrene levels inurine indicate that the presence of MGP tar on soilresults in a considerable decrease in PAH systemicbioavailability (9−75%). Similarly, PAH bioavailabilityestimates based on chemical:DNA adduct formation rangedfrom nondetectable to 76%. These results clearly indicatethat the bioavailability of PAH is less than 100% whensoil contaminated with MGP tar is ingested by mice. Inaddition, the experimental methods employed in this studyappear suitable for evaluating the effects of soil on thegastrointestinal absorption and systemic bioavailability ofPAH from soil containing complex organic mixtures.
